Microelectrode

 

Standard protocols in Purkinje fibres or papillary muscles for GLP studies:

• six (6) preparations (n=6)
• Four (4) cumulative increasing concentrations of assay compound
• Washout period or positive control at the end of the experiment
• Normal [K⁺] ext
• Two stimulation rates: 1 Hz and 0.2 Hz (reverse use dependency)

OPTIONS: External hypokaliemic conditions, mycrosyringe, chronic pre-treatment, incremental stimulation rate.

1. Action potential recordings from rabbit and dog Purkinje fibres

 

Treated groups could be compared to a control vehicle group performed in the same conditions.

2. Action potential recordings from guinea pig papillary muscles

3. Action potential recordings from isolated cardiomyocytes

Current clamp: RP (mV), Amplitude (mV), APD₂₀ (ms), APD₈₀ (ms)
Voltage clamp: specific current would be recorded with specific inhibitors.
